چکیده
The enzyme 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR, EC 1.1.1.34) is a key enzyme in the isoprenoid biosynthetic pathway. We have isolated partial cDNAs from wheat (Triticum aestivum) using the polymerase chain reaction. Comparison of deduced amino acid sequences of these cDNAs shows that they represent a small family of genes that share a high degree of sequence homology among themselves as well as among genes from other organisms including tomato, Arabidopsis, hamster, human, Drosophila, and yeast. Southern blot analysis reveals the presence of at least four genes. Our results concerning the tissue-specific expression as well as developmental regulation of these HMGR cDNAs highlight the important role of this enzyme in the growth and development of wheat.

The distribution of 3-hydroxy-3-methylglutaryl coenzyme A reductase in the small intestine of rats fed raw or gelatinized starch as a carbohydrate source.
The effect of gelatinization of dietary starch (wheat, corn and potato) on the distribution of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase along the small intestine of rats was studied in villus and crypt cell populations isolated by graduated scraping. Gelatinized starch preferentially stimulated jejunal reductase activity in both villus and crypt cells and consequently retained ...
متن کاملOver - expression Effect of Gene Encoding 3-hydroxy-3-Methylglutaryl-CoA Reductase on Production of Taxol in Iranian Hazel (Corylus avellana L.)
Background: Sustainable and commercial production of taxol as an anti cancer drug is a critical point to its clinical application. Nowadays, hazel because of rapid growth and wide range distribution is considered as an alternative source of Taxol. Objective: To increase taxol production the cDNA encoding 3-hydroxy-3-methylglutaryl-CoA reductase (HMGR) from Iranian hazel (GeneBank accession num...
متن کاملSaccharomyces cerevisiae contains two functional genes encoding 3-hydroxy-3-methylglutaryl-coenzyme A reductase.
We have isolated two genes from yeast encoding 3-hydroxy-3-methylglutaryl-coenzyme A reductase [hydroxymethylglutaryl-coenzyme A reductase (NADPH); HMG-CoA reductase; EC 1.1.1.34], the rate-limiting enzyme of sterol biosynthesis. These genes, HMG1 and HMG2, were identified by hybridization to a cDNA clone encoding hamster HMG-CoA reductase. DNA sequence analysis reveals homology between the ami...
متن کاملEssentiality, expression, and characterization of the class II 3-hydroxy-3-methylglutaryl coenzyme A reductase of Staphylococcus aureus.
Sequence comparisons have implied the presence of genes encoding enzymes of the mevalonate pathway for isopentenyl diphosphate biosynthesis in the gram-positive pathogen Staphylococcus aureus. In this study we showed through genetic disruption experiments that mvaA, which encodes a putative class II 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase, is essential for in vitro growth of S...
متن کاملProperties of 3-hydroxy-3-methylglutaryl coenzyme A reductase solubilized from rat liver and hepatoma.
In hepatomas, the activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase, the rate-controlling enzyme in cholesterol biosynthesis, is not normally suppressed by cholesterol. To examine the biochemical mechanism of this loss of feedback control of cholesterol synthesis, a comparison was made of the properties of 3-hydroxy-3-methylglutaryl coenzyme A reductase after solubilization and partia...
